  •   1991年在上海第二医科大学获得博士学位,1994年2月至1999年1月期间,在美国国立卫生研究院(NIH)从事博士后和高级访问学者研究,1998年入选中科院疫学专业。1999年1月至今在中科院上海生物化学与细胞生物学研究所,组建分子免疫室,任课题组组长,博士生导师。2006年4月至2016年12月,任中国科学院上海巴斯德研究所所长。

      孙兵研究员长期从事T辅助细胞(Th)的分化机制研究,并探讨不同的Th细胞亚群和ILC2细胞在炎症和免疫性疾病中的作用机制。现为美国免疫学家学会会员(American Association of Immunologists; AAI),中国免疫学会副理事长。2003年荣获国家杰出青年研究基金支持,2012年荣获“上海市自然科学二等奖”和2013年荣获“国家自然科学二等奖”。

  • Th细胞分化机制和ILC2细胞功能的研究及其在炎症和免疫性疾病中作用机制研究

  •   1. Allergic Asthma

      近年来因为空气污染(如雾霾等)的原因,哮喘发病率持续上升,而现有手段并不能有效控制。研究表明,过敏原可以引起肺粘膜上皮细胞损伤,通过释放IL-25, IL-33和TSLP等炎症性细胞因子,进一步激活II型淋巴样免疫细胞(Innate lymphoid type 2 cells: ILC2)。 ILC2细胞可以分泌IL-5和IL-13,同时在过敏原的作用下,可以驱动幼稚CD4+T细胞分化成Th2细胞,而最终诱导哮喘疾病。实验室长期从事Allergic Asthma疾病分子机制研究,目前重点聚焦Th2和ILC2这两种免疫细胞的功能研究及相互作用的调控机制。利用过敏性哮喘小鼠模型,结合哮喘病人的临床数据,研究新的转录调控因子、表观遗传分子和代谢信号通路及产物如何调节这些细胞的功能及它们对哮喘致病进程的调控作用机制,为开发新药和防治哮喘性疾病提供新思想和新策略。

      2. Cancer Immunotherapy

      自2011年开始,多种免疫检查点阻断剂和靶向免疫细胞药物在多种肿瘤治疗中取得了良好的临床效果,已经发展成为目前公认的最具潜力的癌症治疗策略。已上市的免疫治疗方法,只在部分病人中展现出良好的效果,而在大部分肿瘤患者中出现耐药性和预后差等现象,使肿瘤免疫治疗具有一定的局限性。本研究依托成熟的小鼠实验系统和临床医院提供的病人样本,重点探究Lung Cancer形成过程中CD8+及Treg细胞的重要调控作用,并遴选特异性靶标,运用实验室完善的单细胞技术制备抗体和药物研发技术体系,制备靶向肿瘤Treg和提升CD8+细胞功能,进而开发有抗肿瘤作用的新型抗体,在体内和体外评价新抗体的抗肿瘤效果,开发新药为针对Lung Cancer的抗体免疫治疗提供新方案和新策略。

      3. Antibody drug discovery

      实验室有近20年抗体研究历史,已有完备抗体研究相关平台。实验室已建立了完善的single-cell RT-PCR 制备全人单克隆抗体技术,尤其针对感染性疾病,能够快速获得有治疗性作用的抗体候选药物。同时,针对一些抗原免疫小鼠产生免疫耐受,无法获得单克隆抗体的问题,利用兔的免疫细胞具有体细胞高频突变和体细胞基因置换两种机制,可以打破免疫耐受。实验室建立了基于single-cell RT-PCR 制备兔单克隆抗体技术。我们的技术体系涵盖靶点发现与验证,抗体制备和功能评估以及知识产权管理等抗体药物研发的关键步骤。实验室已成功研发并转化了HCV广谱中和全人抗体等项目。目前实验室正聚焦哮喘及肿瘤等疾病。开展基础和临床研究,研发创新的抗体药物。
